Abstract
Inositol signaling supports the function of serotonin, norepinephrine, and cholinergic receptors, providing a rational basis for anxiolytic effects. However, CANMAT does not recommend it due to the absence of grade A evidence. The main interaction concern is with lithium and valproate, which work by depleting CNS inositol. Watch for theoretical efficacy loss. In bipolar pilot studies, inositol at 4 grams daily preserved mood-stabilizer efficacy while reducing hypothyroidism, weight gain, and metabolic side effects.
