Abstract

<jats:p>Производственно значимые культуры микроорганизмов во многом определяют качество, безопасность и функциональные свойства ферментированных пищевых продуктов. Их оценка традиционно включала подтверждение таксономической принадлежности, отсутствие признаков патогенности и токсигенности, а также проверку технологической пригодности. В последние годы требования к характеристике таких культур расширяются, поскольку безопасность пищевых микроорганизмов все чаще рассматривается в контексте распространения антибиотикорезистентности и концепции «Единое здоровье». Для заквасочных и потенциально пробиотических штаммов особое значение приобретает выявление приобретенных генов антибиотикорезистентности (antibiotic resistance genes, ARG), определение их генетического окружения и оценка связи с мобильными генетическими элементами (mobile genetic elements, MGE), способными участвовать в горизонтальном переносе. Цель обзора заключалась в обобщении современных нормативно-методических и научных подходов к оценке антибиотикорезистентности пищевых микробных культур. Проанализированы публикации и документы, отражающие рекомендации FAO/WHO, систему EFSA/QPS, подходы FDA/GRAS и Health Canada, а также действующие требования ЕАЭС и Российской Федерации. Показано, что наиболее обоснованная схема оценки должна быть штамм-специфической и включать фенотипическую оценку чувствительности к антибиотикам, полногеномное секвенирование, анализ генетического окружения выявленных ARG и таргетный мониторинг производственных партий. Отдельное внимание уделено разграничению природной и приобретенной устойчивости, а также роли плазмид, транспозонов, интегронов и других MGE при интерпретации потенциальной переносимости. Подчеркнуто, что фенотипическая устойчивость без анализа генетического контекста не позволяет достоверно оценить риск передачи детерминант устойчивости. Результаты обзора демонстрируют целесообразность включения ARG/MGE-профиля в систему паспортизации микробных культур как дополнительного критерия их геномной безопасности, прослеживаемости и сопоставимости с международными подходами к оценке пищевых микроорганизмов.</jats:p> <jats:p>Industrially significant microbial cultures largely determine the quality, safety, and functional properties of fermented foods. Traditionally, their assessment has included confirmation of taxonomic identity, absence of pathogenic and toxigenic traits, and evaluation of technological suitability. In recent years, the requirements for the characterization of such cultures have expanded, since the safety of food-associated microorganisms is increasingly considered in the context of antimicrobial resistance dissemination and the One Health concept. For starter and potentially probiotic strains, particular importance is attached to the detection of acquired antibiotic resistance genes (ARG), determination of their genetic context, and assessment of their association with mobile genetic elements (MGE) capable of participating in horizontal gene transfer. The aim of this review was to summarize current regulatory, methodological, and scientific approaches to assessing antimicrobial resistance in food microbial cultures. Publications and documents reflecting FAO/WHO recommendations, the EFSA/QPS system, FDA/GRAS and Health Canada approaches, as well as current requirements of the EAEU and the Russian Federation, were analyzed. It is shown that the most scientifically justified assessment scheme should be strain-specific and include phenotypic antimicrobial susceptibility testing, whole-genome sequencing, analysis of the genetic context of detected ARG, and targeted monitoring of production batches. Special attention is given to distinguishing intrinsic and acquired resistance, as well as to the role of plasmids, transposons, integrons, and other MGE in the interpretation of potential transferability. It is emphasized that phenotypic resistance without analysis of the genetic context does not allow a reliable assessment of the risk of resistance determinant transfer. The results of the review demonstrate the feasibility of including ARG/MGE profiling in the passporting system for microbial cultures as an additional criterion of their genomic safety, traceability, and comparability with international approaches to the assessment of food-associated microorganisms.</jats:p>
