Abstract

<title>Abstract</title> <p> This study investigates the isolation and characterization of polyethylene terephthalate (PET)-degrading bacteria from plastic-contaminated soils in Western India. Soil samples were collected from landfill sites and screened using PET-based media to identify isolates capable of polymer degradation. A total of 25 bacterial isolates were obtained and evaluated based on hydrolysis zone formation and growth under varying conditions. Selected isolates demonstrated significant PET degradation, including tolerance to high salt concentrations. Biochemical characterization and 16S rRNA sequencing identified dominant strains belonging to genera <italic>Pseudomonas</italic> and <italic>Stutzerimonas</italic> . Thin layer chromatography confirmed degradation intermediates such as terephthalic acid and bis(2-hydroxyethyl) terephthalate. These findings highlight the potential of native microbial communities for sustainable plastic waste bioremediation. <bold>Importance</bold> : This study is significant as it demonstrates a potential approach to address plastic pollution. Researchers found bacteria at waste sites that can break down plastic into simpler substances. This means burning or dumping plastic, which hurts the environment, could be avoided. These bacteria may be applied for plastic bioremediation. The study also found that some bacteria can work in high saline conditions, which is useful for cleaning up plastic pollution in oceans. This research is a step towards better management of plastic waste. </p>
