Abstract

<title>Abstract</title> <p>Two recombinant protein vaccines (Arexvy and Abrysvo) and one mRNA vaccine (mResvia) licensed by the Food and Drug Administration (FDA) were derived from the prefusion F protein. In this study, we screened out a new recombinant protein vaccine based on prefusion and trimeric RSV F conformation. This protein with two introduced disulfide-linkage pairs and three cavity-filling mutations exhibited admirable stability when kept at different temperatures for quite a long time. Our experiments revealed that with Al(OH)3, it generated robust immunogenic responses in mice as well as RSV-naïve and RSV-experienced cotton rats, and offered sufficient protection against RSV infection without a polarization of Th2-associated cytokines or increasing lung inflammation. Due to its stability when formulated with Al(OH)3, the formulation could be a liquid vaccine providing a viable option for RSV-vulnerable groups.</p>
